Characteristic findings

The cytoplasm of mast cells contains many granules that are strongly stained by platinum blue. Consequently, these cells appear dark blue in light-microscope images, but are strikingly bright and clear under a LVSEM 3), 4). The three-dimensional shape of each granule in the cells becomes more evident when the cells are examined with a LVSEM at higher magnification. Even the size of the granules can be determined using the measurement mode of the LVSEM.

Observations

Mast cells appear dark blue in light-microscope Fig. 000004a, whereas they are extremely bright and prominent under a LVSEM. In higher-magnification LVSEM images, the mast cell cytoplasm filled with many granules appears three-dimensional.
